About 300 research groups benefit from our support with main domains of application in life science, biomedicine, physics, chemistry, and economy. In addition, sciCORE is one of the infrastructure nodes of the national initiatives BioMedIT and Swiss Personalized Health Network, providing a safe environment for research based on patient data. This position will be based in the Department of Biomedicine (https://biomedizin.unibas.ch/en/) in Basel, Switzerland - one of the world's leading locations for life sciences, home to several companies and institutions.Prof. Beltra's group is studying the factors governing T-cell differentiation programs in settings of viral infections and cancers. Notably, we are particularly interested in identifying new pathways regulating the process of CD8 T cell exhaustion, an hypofunctional program acquired by CD8 T cells exposed to persistent inflammation and antigenic stimulation. Indeed, CD8 T cell exhaustion represents a central limitation to current anti-cancer immunotherapies and strategies aiming at reversing this process are still lacking. By exploring the intimate mechanisms regulating CD8 T cell exhaustion, our group aims to identify innovative opportunities to reverse this process and unleash the full potential of current cancer immunotherapies (e.g. PD-1-based therapy). PhD student position: Biochemistry/Molecular Toxicology(100% / available immediately)The Division of Molecular and Systems Toxicology at the University of Basel is offering a PhD student position to investigate substances altering corticosteroid hormone action.Your positionCorticosteroids regulate essential physiological processes. Substances altering their production, cell-specific metabolism or receptor-mediated signalling can aggravate various diseases including inflammatory, cardio-metabolic as well as developmental and reproductive disorders. Despite the high interest in identifying endocrine disrupting chemicals (EDCs), the identification and characterization of substances disturbing corticosteroid action and elucidation of their mechanism-of-action is a largely neglected topic.The present project is a collaborative effort with four other partners to study chemical-induced steroid perturbations. The project is financed by the Swiss Centre for Applied Human Toxicology. The aim of the project is to establish and apply methods and cell-based assays to assess chemical-induced alterations in glucocorticoid pre-receptor enzyme activity and regulation. Promoter-reporter cell lines will be generated for HSD11B1, converting inactive cortisone to the potent glucocorticoid cortisol, and HSD11B2, catalysing the reverse reaction. Enzyme activity assays that are already established will be applied to test for direct inhibitory activity of compounds. Cell lines stably expressing either these promoter-reporter constructs or glucocorticoid receptor (GR) reporter plasmids will be constructed. Cell lines of interest include placental JEG3, colon SW620, and renal RCCD2 for HSD11B2, and hepatic and macrophage cell lines for HSD11B1. Compounds that will be included in the analyses comprise PFAS, phthalates, bisphenols, UV-filters and parabens. Depending on activity and relevance from literature, the most promising active compound(s) can be further investigated in additional cell models or in animals. Methods applied include cell culture, transfection and gene expression analyses, molecular cloning, receptor function analyses, and enzyme activity measurements. Resistance to endogenous and pharmacological glucocorticoids is a major problem. The proposed project investigates endogenous and exogenous substances that influence glucocorticoid sensitivity. In collaboration with rheumatologists, we will analyze steroid profiles in patients with rheumatic diseases, attempting to identify markers indicating glucocorticoid resistance. Using macrophage models, the impact of endogenous and exogenous substances on corticosteroid receptor sensitivity and cytokine production will be studied.The successful candidate will collaborate with our senior analytical scientist to determine/analyze steroid profiles from patients with rheumatic diseases after treatment with glucocorticoids. In collaboration with rheumatologists, the candidate is expected to perform data analyses, combining steroid concentrations and clinical data.In addition, a main part of the project includes the establishment and use of protocols with macrophages for the investigations of endogenous and exogenous substances that alter the sensitivity of glucocorticoid and mineralocorticoid receptors for their ligands. Notably, we are particularly interested in identifying new pathways regulating the process of CD8 T cell exhaustion, an hypofunctional program acquired by CD8 T cells exposed to persistent inflammation and antigenic stimulation. Indeed, CD8 T cell exhaustion represents a central limitation to current anti-cancer immunotherapies and strategies aiming at reversing this process are still lacking. By exploring the intimate mechanisms regulating CD8 T cell exhaustion, our group aims to identify innovative opportunities to reverse this process and unleash the full potential of current cancer immunotherapies (e.g. PD-1-based therapy). To this end, we have developed relevant murine models for the study of CD8 T cell-exhaustion and are leveraging cutting-edge technologies (single cell omics, ATACseq) and innovative immunological and genetic perturbation tools (Retroviruses, CRISPR), to identify and characterize new pathways of interest for a better therapeutic manipulation of exhausted CD8 T cells. Mai 2024 oder per Vereinbarung eine*n Doktorand*in im Fachbereich Recht.Viele bilaterale Abkommen zwischen der Schweiz und der Europäischen Union (EU) sehen Rechte für Einzelpersonen vor, seien es Menschen oder z. B. Unternehmen. In der Praxis hängt die Stärke dieser Rechte von den Möglichkeiten ihrer Durchsetzung ab, u.a. im Streitbeilegungsverfahren zwischen den Parteien der Abkommen. Das bilaterale Recht Schweiz-EU ist diesbezüglich im Vergleich zum EU- und zum EWR-Recht deutlich schwächer ausgestaltet. Die Schweiz und die EU arbeiten seit einigen Jahren an der Modernisierung der institutionellen Regeln für wichtige Abkommen. Im Rahmen der Forschungsarbeit soll das Durchsetzungssystem der bilateralen Kernabkommen im Hinblick auf die Durchsetzung mit anderen Aussenabkommen der EU (u.a. EWR, Ankara-Abkommen, Ukraine-Abkommen, Brexit-Austrittsabkommen) verglichen werden. The PhD-positions are part of the recently funded SNSF Sinergia Project ?Holocene hydroclimate, drought dynamics and environmental changes recorded in multiple archives from SW Asia (MITRA)".Your positionOur knowledge of climate variability in SW Asia remained fragmentary due to scarcity of precisely-dated and highly-resolved paleoclimate and paleoenvironmental reconstructions. One of the main goals of SNSF Sinergia project MITRA is to develop such records. Both PhD-students will investigate speleothems (stalagmites and flowstones) from several caves in Turkey, Iraq and Iran to reconstruct hydroclimate variability during the Holocene period. This is achieved by using a wide range of geochemical methods, such as Uranium-series dating, stable isotope and trace element analysis. The speleothem-based reconstructions will be then compared to lake and marine records and regional climate model simulations, which will be developed by MITRA co-investigators Professor Christoph Raible (University of Bern) and Dr. Morteza Djamali (CNRS Marseille) and other MITRA members.Your profileWe are looking for highly-motivated and proactive candidates with a master degree in geosciences or environmental sciences. Das CEPS wurde 2008 gegründet und verbindet international anerkannte Forschung mit der praktischen Umsetzung in Weiterbildungsangeboten für den Nonprofit-Sektor. Zur Stärkung seiner Forschungstätigkeit sucht das CEPS nach Vereinbarung eine wissenschaftliche Mitarbeiterin / einen wissenschaftlichen Mitarbeiter angewandte Forschung (60-80 %).Ihre AufgabenIn Ihrer Funktion entwickeln und realisieren Sie Projekte in der angewandten Forschung wie Studien, Kurzberichte und Auftragsforschung.
